ExoSAP-IT® is added directly to a PCR product with one pipetting step. Applications such as DNA sequencing, SNP or SBE analysis only require a small portion of the PCR product for analysis. It's ideal for treating these samples with no need to purify the entire PCR mixture.

 
The ExoSAP-IT® method is designed to require a minimum of “hands-on” time and is completely amenable to medium or high throughput applications. Only simple pipette transfers are required, therefore, many  samples can be processed at once in multiple tubes or plates, either manually or by robotics.
 
ExoSAP-IT® is an enzymatic method that degrades excess primers and dephosphorylates dNTPs left over in a PCR mixture. A simple pipetting step is all that is required. ExoSAP-IT® treatment avoids variables that can be introduced with multiple step protocols and columns.
 
The key to ExoSAP-IT® is that it enzymatically treats PCR products  to prevent unused primers & dNTPs from interfering in downstream applications. Samples are not exposed to columns or other methods that have inherent sample loss. Short and long PCR products can be ExoSAP-IT®-treated with absolutely no sample loss.
 


A 15 minute incubation at 37°C is all that is needed for ExoSAP-IT® to enzymatically clean up a PCR mixture. No buffer exchange is required. Following clean-up, ExoSAP-IT® is inactivated simply by heating to 80°C for  15 minutes. Within just 30 minutes, the PCR product is ready for immediate analysis.
